SBA-15 mesoporöse Silikananotherapeutika

 

Das hier präsentierte SBA-15 ist ein mesoporöses Silikat mit hoher spezifischer Oberfläche (>800 m2/g), welches wir erfolgreich erforschen für die Antikrebstherapie. Bekannte Wirkstoffe (Cisplatin), neue metallorganische Verbindungen (basierend auf Titan, Zinn, Ruthenium, Platin) oder Naturwirkstoffe konnten eingelagert werden und zeigten erfolgreiche, hohe Wirksamkeit an verschiedenen Krebszelllinien und deutlich geringere Toxizität in vivo.

Krebszellen nehmen die einzelnen Nanopartikel mit einer Größe von < 1μm auf und der Wirkstoff kann an Ort und Stelle wirken ohne vorher Nebenwirkungen zu verursachen. Zum ersten Mal wurde mit metallbasierten Medikamenten eine Differenzierung und Umprogrammierung von überlebenden Klonen der Tumorzellen zu seneszenten Zellen beobachtet.

Der Weg von Träger und Wirkstoff konnte bereits mittels fluoreszenzmarkierten Verbindungen verfolgt werden. Die Forschung der Arbeitsgruppe unter Prof. Kaluđerović beschäftigt sich mit Wirkstoffen, die besonders aktiv am SBA-15 sind. Das Ziel sind neue Leitstrukturen, die außerhalb des Trägers geringe Toxizität zeigen, aber zusammen mit SBA-15 Synergieeffekte aufweisen. Somit können mit einfachen Mitteln Nanopartikel als Träger als auch Wirkstoffe hergestellt werden, die Tumorzellen nicht nur absterben lassen mittels Apoptose, sondern  auch das Wachstum verbleibender Krebszellen stoppen anstatt es zu stimulieren und Resistenzen überwinden können durch Applikation in diesem mesoporösem Träger.

SBA-15 is a mesoporous silica with high specific surface (>800 m2/g), which is used in our research upon therapeutics for antitumoral agents. Already used agents like cisplatin and new metal-organic compounds (based on Ti, Sn, Pt, Ru) as well as natural compounds can and were adsorbed into the SBA-15 pores and proofed to be efficacious vs different cancer cell lines. Reduced toxicity was shown in vivo together with heavenly suppressed tumor growth.

Cancer cells ingest the nanoparticles (e.g. endocytosis), which have a size of less than 1μm and the drug can be released at the target site without interacting with blood plasm, proteins etc. and reducing this way side effects, off-target toxicity and preliminary decomposition. For the first time, metal-based drugs showed differentiation to senescent cells upon treatment in our studies and therefore establishing a new way to diminish tumor growth.

The mechanism of action and the pathway of nanoparticles and drugs were studied with fluorescent markers. Thus, drug release and behavior of SBA-15 can be monitored in vitro. Further research is going on to find new leads, which are especially active adsorbed to silica, but less active and less toxic while acting as free drug. The synergetic effects will make this approach safer and biologically more active at the right targets.

Hence, within this project nanoparticles and drug candidates are produced easily and act together to suppress tumors not just by destroying the cell or activating programmed cell death, but stopping the aggressive tumor growth and surpassing resistances.
